This webinar provides an overview of Antibody-Drug Conjugates (ADCs) and expanded drug conjugates (XDCs), with a focus on Biointron's experiences and capabilities.
The talk will begin with the history and development of ADCs, listing clinically approved ADC products from 2000 to 2024, detailing their targets, payloads, and conjugation strategies. It highlights that interchain cysteine conjugation is currently the dominant method. The presentation then shifts to emerging trends in XDCs, noting a significant increase in interest in novel formats like bispecific ADCs, radioconjugates, and dual-payload ADCs.
Secondly, the presentations will showcase Biointron's extensive case studies in XDC development. These include examples of conjugating various macromolecules (monoclonal antibodies, bispecific antibodies, VHH-Fc, etc.) with different payloads (e.g., MMAE, Dxd) using diverse methods (cysteine conjugation, engineered cysteine, MTG, UAA insertion). The case studies present detailed DAR (Drug-to-Antibody Ratio) analyses via LC-MS. The scope of XDC is further expanded to include other modalities like peptide-drug conjugates, AOCs (Antibody-Oligonucleotide Conjugates), and Nanobody-LNP complexes for in vivo CAR-T applications.
Finally, the talk will introduce Biointron as a company specializing in antibody discovery and production since 2012. It outlines our end-to-end service platform, which includes antibody discovery, optimization, expression, and XDC development/analysis, supported by high-throughput production facilities and an in-house alpaca farm for VHH generation.

Dr. Brady Wu is a multidisciplinary scientist and biotech leader. He holds a Ph.D. in Chemistry from The Ohio State University (2007–2013), where he pioneered solid-state NMR studies of amyloid fibril dynamics, and a Peking University honors B.S. in Biology (2003–2007). At GenScript (2013–2021), he led 100+ teams in recombinant protein production service and global project management. As the Founding CEO of Solvo Biotherapeutics (2022–2024), he built China’s top cell-free protein synthesis platform with a 15+ team. Now leading the Protein Science group at Biointron (2024–present), he drives ADC conjugation, developability assessment, and recombinant protein/antibody R&D efforts in the company.
